Anbalcabtagene autoleucel (Anbal-cel) is a next-generation autologous CD19-directed chimeric antigen receptor (CAR)-T cell therapy developed using Curocell’s OVIS platform. It uniquely incorporates knock-downs of both PD-1 (programmed cell death protein 1) and TIGIT (T cell immunoglobulin and ITIM domains), which are immune checkpoint receptors implicated in T cell exhaustion. By reducing the expression of these inhibitory molecules, Anbal-cel aims to enhance CAR-T cell persistence and antitumor efficacy versus conventional anti-CD19 CAR-T therapies. It has demonstrated superior eradication of CD19-positive tumor cells in preclinical studies, and early clinical trials show promising efficacy and a tolerable safety profile. Its primary development indications include relapsed/refractory large B-cell lymphoma, follicular lymphoma, central nervous system lymphoma, and high-grade B-cell lymphoma[1][5][9].
