Anbenitamab is a bispecific monoclonal antibody engineered to target two distinct extracellular domains (II and IV) of the human HER2 receptor. It is derived from trastuzumab and pertuzumab, enabling simultaneous binding to different HER2 epitopes. This dual-binding mechanism blocks both ligand-dependent and ligand-independent HER2 signaling pathways, inhibits heterodimerization, and disrupts downstream signaling crucial for tumor cell proliferation and survival. Anbenitamab also mediates antibody-dependent cellular cytotoxicity (ADCC), leading to apoptosis in HER2-overexpressing tumor cells. Developed using CRIB (Charge Repulsion Induced Bispecific) technology, it has demonstrated higher binding affinity than existing HER2-targeted therapies such as trastuzumab plus pertuzumab, with efficacy even in tumors expressing low or medium levels of HER2 or those resistant to trastuzumab. Its primary indications are for the treatment of various cancers characterized by overexpression of the HER2 receptor, including breast cancer and gastric cancer[1][4][5][7].
