ANC-1 (antibody nanoconjugate-1) is a preclinical-stage antibody-targeted nanoparticle (immunoliposome) developed for the treatment of HER2-positive breast cancer. It consists of the chemotherapeutic agent docetaxel encapsulated within a pegylated (stealth) liposome, which is covalently engrafted with the anti-HER2 monoclonal antibody trastuzumab on its surface. This design is intended to improve the therapeutic index of the docetaxel-trastuzumab combination by enhancing drug trafficking, tumor-specific delivery, and cellular uptake via receptor-mediated endocytosis, thereby reducing systemic toxicities associated with the standard doublet therapy.
