ANC-2 (antibody nanoconjugate-2) is a preclinical-stage antibody-nanoconjugate (specifically, an immunoliposome) developed for the treatment of HER2-positive breast cancer. It consists of the cytotoxic taxane chemotherapy docetaxel encapsulated within a pegylated stealth liposome made of synthetic lipids (such as DOTAP and PC), which is covalently engrafted with the anti-HER2 monoclonal antibody trastuzumab via a maleimide linker. Trastuzumab serves as both a therapeutic agent and a targeting ligand to direct the nanoparticle to HER2-expressing cancer cells, facilitating receptor-mediated endocytosis and intracellular delivery of docetaxel. In preclinical studies, ANC-2 demonstrated significant in vitro antiproliferative activity against human breast cancer cell lines, although its counterpart made of natural lipids (ANC-1) showed superior stability and encapsulation efficiency.
