Overview

LYS228 (ancremonam) is a novel intravenous monobactam antibiotic developed to treat infections caused by multidrug-resistant Gram-negative bacteria, particularly Enterobacteriaceae expressing extended-spectrum β-lactamases and carbapenemases. Its mechanism of action involves inhibition of bacterial cell wall synthesis through covalent modification of penicillin binding protein 3 (PBP3), leading to disruption of peptidoglycan synthesis and cell division. LYS228 is notable for its stability against both metallo-β-lactamases (MBLs) and most serine β-lactamases (SBLs), addressing resistance mechanisms that limit the effectiveness of other β-lactams. The drug was initially discovered by Novartis and later licensed to Boston Pharmaceuticals for further development. It has been evaluated in phase 2 clinical trials for complicated urinary tract infections and complicated intra-abdominal infections in adults[1][4][5][6][8].
