This drug is a **combination therapy** consisting of **andecaliximab**, a monoclonal antibody that selectively inhibits matrix metalloproteinase-9 (MMP9), and **nivolumab**, a human IgG4 monoclonal antibody that inhibits programmed death-1 (PD-1) on T cells. Andecaliximab disrupts tumor-promoting processes in the extracellular matrix and tumor microenvironment by blocking MMP9. Nivolumab blocks the PD-1 receptor, thereby enhancing T-cell-mediated immune responses and promoting anti-tumor immunity. This combination has been investigated mainly for the treatment of advanced gastric or gastroesophageal junction adenocarcinoma; although it demonstrates a manageable safety profile, it did not significantly improve outcomes compared with nivolumab monotherapy in randomized clinical trials[1][3][7].
