Andes-1537 is a proprietary short single-stranded phosphorothioate deoxyoligonucleotide antisense oligonucleotide developed by Andes Biotechnologies. It targets a novel non-coding mitochondrial RNA (ncmtRNA), specifically the Antisense non-coding mitochondrial RNA (ASncmtRNA). By base pairing with this target, Andes-1537 forms an RNA-DNA hybrid that is hydrolyzed by RNase H and DICER, resulting in microRNAs. This process leads to gene silencing effects in cancer cells, including induction of apoptosis through downregulation of anti-apoptotic proteins like survivin, inhibition of proliferative signaling via suppression of cyclin D1 and cyclin B1 expression, and reduction in metastatic potential by inhibiting proteins such as n-cadherin and β-catenin. The drug has shown preliminary efficacy signals in hard-to-treat cancers such as pancreatic cancer and cholangiocarcinoma during phase 1 trials[2][3][5].
