Overview

Anemoside B4 is a pentacyclic triterpenoid saponin isolated predominantly from the roots of *Pulsatilla chinensis*, a traditional Chinese medicinal herb. It exhibits potent anti-inflammatory and immunomodulatory activities, demonstrated in various preclinical studies. Mechanistically, Anemoside B4 inhibits the activation of multiple pro-inflammatory pathways, including the suppression of pro-inflammatory cytokine release (TNF-α, IL-1β, IL-6) and reduction of neutrophil and white blood cell counts in models of infectious pneumonia. It acts via inhibition of the toll-like receptor 4 (TLR4)/MyD88 signaling pathway, directly interacts with and inhibits NEK7, thereby blocking NLRP3 inflammasome activation, and is also a direct inhibitor of pyruvate carboxylase, affecting cellular metabolic reprogramming and inflammation. Anemoside B4 is also able to inhibit E-selectin secretion and regulate additional inflammatory and immune pathways. It is being investigated as a therapeutic candidate for bacterial and viral pneumonia, acute gouty arthritis, inflammatory bowel disease/colitis, and other immunoinflammatory disorders[1][3][5][4][7][8][9].
