Anetumab ravtansine is an investigational antibody-drug conjugate (ADC) composed of a fully human anti-mesothelin monoclonal antibody linked to the cytotoxic maytansinoid tubulin inhibitor DM4. The monoclonal antibody component specifically targets mesothelin, a tumor-associated antigen highly expressed in several cancers, including ovarian cancer and mesothelioma. Upon binding to mesothelin-expressing tumor cells, the ADC is internalized, releasing DM4 intracellularly where it binds to tubulin and disrupts microtubule assembly/disassembly dynamics. This results in mitotic arrest and apoptosis of the targeted cancer cells. Anetumab ravtansine was developed primarily for solid tumors expressing mesothelin but has been discontinued for all indications after reaching various stages of clinical development[1][2][5][6].
