**Ang1-C4bp** is a recombinant heptameric fusion protein combining the fibrinogen-like domain (FLD) of angiopoietin-1 (Ang1) with the C-terminal oligomerization scaffold of complement C4-binding protein (C4BP), designed to mimic native Ang1's multimeric structure for potent Tie2 receptor activation. It promotes Tie2 clustering on endothelial cells, enhancing downstream signaling via AKT phosphorylation, which stabilizes vasculature, reduces vascular leakage, and protects against permeability induced by VEGF or inflammation. Developed as an injectable biologic for vascular disorders, it demonstrates superior stability, systemic efficacy, and activity in preclinical models including HUVEC cells, mice, and rabbits, with intravitreal pharmacokinetics supporting ocular applications.
