ANG1005 + bevacizumab is a combination regimen consisting of **ANG1005 (paclitaxel trevatide)** and **bevacizumab**, explored mainly for the treatment of high-grade gliomas, particularly in populations that have failed or are refractory to prior therapies, including bevacizumab itself. ANG1005 is a novel peptide-drug conjugate that links three paclitaxel molecules to Angiopep-2, which targets the low-density lipoprotein receptor-related protein-1 (LRP1), facilitating transport across the blood-brain barrier (BBB) and into brain parenchyma and tumor cells. ANG1005 acts by stabilizing microtubules, leading to inhibition of cell proliferation and apoptosis, similarly to paclitaxel but with enhanced CNS penetration. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting tumor angiogenesis. This combination targets both tumor cell proliferation (via microtubule stabilization and delivery across the BBB) and tumor vascular supply (by VEGF inhibition). Clinical studies have focused on use in recurrent high-grade glioma, including bevacizumab-refractory populations[1][2][3][4].
