ANG2002 is a preclinical peptide-drug conjugate (PDC) developed by Angiochem for the treatment of chronic pain and other central nervous system (CNS) disorders. It leverages Angiochem's proprietary Angiopep technology, which utilizes a peptide (Angiopep-2) derived from the Kunitz domain to cross the blood-brain barrier (BBB) via receptor-mediated transcytosis targeting the low-density lipoprotein receptor-related protein 1 (LRP1). The therapeutic payload of ANG2002 is a neurotensin analog designed to activate neurotensin receptors (NTR) within the brain. Neurotensin is an endogenous tridecapeptide that functions as both a neurotransmitter and a neuromodulator, with potent analgesic properties when acting centrally. By facilitating the delivery of neurotensin across the BBB, ANG2002 aims to provide a novel approach to managing chronic pain while avoiding the limitations of poor CNS penetration and rapid systemic degradation typically associated with native neurotensin.
