Overview

Angiocidin is a 41 kDa recombinant human protein that targets the TNF superfamily death receptor 6 (DR6, also known as TNFRSF21). Binding to DR6, angiocidin triggers differentiation of leukemic cells—specifically, inducing terminal differentiation of acute myeloid leukemia (AML) cells—and activates the host immune system, including conversion of monocytes to activated macrophages and T-cell activation via cytokine release. In preclinical models, angiocidin also inhibits tumor proliferation and angiogenesis, including direct inhibition of breast cancer cell growth (via EGFR and NF-κB pathways activation), and induces apoptosis in endothelial cells by inhibiting proteasome activity through polyubiquitin binding. Its biological activities have included tumor growth suppression, increased survival in animal models, and potent synergy with standard AML chemotherapies. Angiocidin is under development primarily as a therapy for AML, but has also been tested in preclinical settings for solid tumors such as breast cancer[1][2][3][4][5][6][7][8][9].
