Angiopep-2-conjugated silver nanoparticles are a targeted nanotherapeutic system designed to cross the blood-brain barrier (BBB) and deliver therapeutic agents to the central nervous system. The construct consists of a silver nanoparticle core functionalized with Angiopep-2, a 19-amino acid peptide derived from the Kunitz domain of aprotinin. Angiopep-2 acts as a ligand for the low-density lipoprotein receptor-related protein 1 (LRP1), which is highly expressed on the endothelial cells of the BBB and on various brain tumor cells, such as glioblastoma. By utilizing receptor-mediated transcytosis, these nanoparticles can effectively bypass the BBB. Once in the brain parenchyma, the silver nanoparticles exert intrinsic cytotoxic effects through the generation of reactive oxygen species (ROS) and induction of apoptosis, making them candidates for the treatment of primary brain tumors and brain metastases.
