Overview

Angiotensin 1-7 (Ang 1-7) is a naturally occurring biologic peptide and a key component of the alternative renin-angiotensin system (RAS) pathway, acting as the primary endogenous ligand for the Mas receptor (MasR). Developed as a therapeutic candidate by researchers at the University of Arizona, Angiotensin 1-7 is being investigated for its neuroprotective and anti-inflammatory properties. Its mechanism of action involves the activation of the Mas receptor, which leads to the inhibition of microglial activation, reduction of pro-inflammatory brain cytokines, and suppression of vascular endothelial reactive oxygen species production. The drug is currently in Phase 1/2 clinical development for the treatment of vascular cognitive impairment and dementia (VCID) and Alzheimer's disease-related dementia (ADRD), specifically targeting heart failure patients who are at high risk for cognitive decline. Additionally, the University of Arizona has developed synthetic glycopeptide derivatives, such as PNA5, to improve the peptide's pharmacological profile, including enhanced brain penetration and extended half-life.
