ANGPTL4 blocking antibody refers to a class of monoclonal antibodies designed to inhibit Angiopoietin-like 4 (ANGPTL4), a secreted protein involved in lipid metabolism, glucose homeostasis, and angiogenesis. In the context of oncology, particularly clear cell renal cell carcinoma (ccRCC), ANGPTL4 is often overexpressed and acts as a pro-angiogenic factor that promotes tumor progression and resistance to standard therapies like tyrosine kinase inhibitors (TKIs). These antibodies work by neutralizing ANGPTL4, specifically targeting its C-terminal domain (cANGPTL4) to prevent its interaction with endothelial cell surface receptors such as the endothelial cell surface chemotaxis receptor (ECSCR). Preclinical research, including studies conducted at the University of Florida, has shown that antibody-mediated inhibition of ANGPTL4 can significantly reduce tumor growth, decrease pathological angiogenesis, and promote vessel normalization, potentially enhancing the efficacy of immune checkpoint inhibitors.
