ANGPTL8 antisense oligonucleotide (Angptl8-ASO) is a second-generation antisense oligonucleotide (ASO) research compound designed to target and silence the mRNA of Angiopoietin-like 8 (ANGPTL8). ANGPTL8 is an insulin-regulated protein that acts as an inhibitor of lipoprotein lipase (LPL), particularly in adipose tissue. By reducing ANGPTL8 levels, the ASO disinhibits LPL activity, which promotes the uptake of circulating triglycerides into adipose tissue and reduces ectopic lipid deposition in the liver. Preclinical studies in rodent models have shown that Angptl8-ASO treatment can significantly reduce hepatic steatosis and improve systemic insulin sensitivity. Additionally, emerging research indicates that ANGPTL8 inhibition may protect against endothelial senescence and angiogenic dysfunction in type 2 diabetes by modulating the RTN4R-PI3K/AKT/FOXO3A signaling pathway, suggesting potential applications in treating diabetic vasculopathy and atherosclerosis.
