Overview

Anisindione is a synthetic anticoagulant and an indanedione derivative, primarily used for the prevention and treatment of venous thrombosis and its extension, atrial fibrillation with embolization, pulmonary embolism, and as an adjunct in the treatment of coronary occlusion. Its mechanism of action involves inhibition of vitamin K–mediated gamma-carboxylation of precursor proteins in the liver. This prevents the formation of active procoagulation factors II, VII, IX, and X as well as anticoagulant proteins C and S. Anisindione does not dissolve existing clots but may limit their extension or prevent secondary thromboembolic complications. It is typically prescribed only if coumarin-type anticoagulants (such as warfarin) cannot be used due to its potent effects and risk profile[1][2][3][5][7].
