Overview

This drug is a combination immunotherapy comprising three agents: **Anktiva (nogapendekin alfa inbakicept-pmln, also called N-803)**, a first-in-class interleukin-15 (IL-15) agonist IgG1 fusion complex; **PD-L1 t-haNK**, an allogeneic natural killer (NK) cell therapy genetically engineered to express a PD-L1-targeted CAR and high-affinity CD16 for antibody-dependent cellular cytotoxicity; and **bevacizumab**, a recombinant humanized monoclonal antibody targeting vascular endothelial growth factor A (VEGFA). - **Anktiva** stimulates and proliferates NK cells and CD4+/CD8+ T cells, enhancing antitumor immune responses and immune memory, overcoming tumor immune evasion, and reversing lymphopenia following chemotherapy or radiotherapy[1]. - **PD-L1 t-haNK** directly targets PD-L1-expressing tumor and myeloid-derived suppressor cells using CAR-NK cell-engineering, providing both direct antitumor cytotoxicity and immune modulation[1][3][5]. - **Bevacizumab** inhibits angiogenesis by selectively neutralizing VEGFA, suppressing new tumor blood vessel formation, normalizing vasculature, and potentially exerting direct cytotoxic effects on VEGFA-expressing tumor cells[2][4]. This combination is under phase 2 clinical investigation for recurrent or progressive glioblastoma and has Regenerative Medicine Advanced Therapy (RMAT) designation in the U.S. for reversal of lymphopenia and multiply relapsed locally advanced or metastatic pancreatic cancer[1][5].
