This four-drug combination regimen is being investigated as a first-line treatment for advanced biliary tract cancer (BTC). It consists of **anlotinib**, a multitarget tyrosine kinase inhibitor (TKI) that inhibits angiogenesis by targeting VEGFR1-3, FGFR1-4, and PDGFRα/β; **bemozulimab** (TQB2450), a humanized monoclonal antibody targeting the PD-L1 immune checkpoint; and the chemotherapy agents **nab-paclitaxel** (albumin-bound paclitaxel) and **cisplatin**. The rationale for this combination is to leverage the synergistic effects of anti-angiogenic therapy, immune checkpoint blockade, and cytotoxic chemotherapy to overcome the limited efficacy of standard treatments in advanced BTC. A Phase 2 clinical trial (NCT05812430) is currently evaluating the safety and efficacy of this regimen in patients with intrahepatic cholangiocarcinoma, extrahepatic cholangiocarcinoma, and gallbladder cancer.
