Overview

Anlotinib + icotinib is a combination therapy used primarily as a first-line treatment for patients with advanced non-small cell lung cancer (NSCLC) harboring EGFR-sensitizing mutations. Anlotinib is a novel, orally administered small molecule multi-target tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR1/2/3), fibroblast growth factor receptors (FGFR1–4), platelet-derived growth factor receptors (PDGFRα/β), and c-Kit, thereby inhibiting tumor angiogenesis and proliferation. Icotinib is a first-generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR TKI) that blocks EGFR signaling to suppress tumor cell proliferation. The combination has demonstrated efficacy and tolerability in clinical trials, improving progression-free survival in patients with EGFR-mutated NSCLC, including those with concurrent pathogenic mutations[1][2][4][5][6].
