Overview

Combination therapy of **anlotinib** (a multitargeted small molecule tyrosine kinase inhibitor with antiangiogenic and antiproliferative activity) and **paclitaxel** (a microtubule stabilizer and widely used cytotoxic chemotherapeutic, often in the albumin-bound, nab-paclitaxel, formulation). Anlotinib inhibits multiple targets, including VEGFR, PDGFR, and FGFR, suppressing tumor angiogenesis and proliferation, while paclitaxel interferes with microtubule disassembly, inhibiting cell division. This combination is under investigation primarily for treatment of advanced or stage IV **non-small cell lung cancer (NSCLC)**, and in some protocols, for other advanced cancers such as esophageal squamous cell carcinoma (ESCC) and small-cell lung cancer (SCLC) with brain metastases. Clinical data show the combination improves response rates, disease control, immune function, and quality of life in advanced NSCLC, and has a manageable safety profile[1][2][3][4].
