Overview
Anlotinib + penpulimab + chemotherapy is a combination regimen under clinical investigation for the treatment of various advanced cancers, including esophageal squamous cell carcinoma (ESCC), non-small cell lung cancer (NSCLC), and other solid tumors. Anlotinib is a small molecule multi-target tyrosine kinase inhibitor that primarily inhibits VEGFR1–3, FGFR, PDGFR, and c-Kit, thereby blocking angiogenesis and tumor proliferation. Penpulimab is a humanized IgG1 monoclonal antibody targeting programmed death 1 (PD-1) receptor on T cells; it blocks the interaction between PD-1 and its ligands to enhance anti-tumor immune responses. The chemotherapy component varies by indication but commonly includes agents such as nab-paclitaxel or paclitaxel with carboplatin. This combination leverages antiangiogenic therapy, immune checkpoint inhibition, and cytotoxic chemotherapy to achieve synergistic antitumor effects[2][5].
