Overview

This is a combination regimen consisting of three agents: anlotinib, toripalimab, and nab-paclitaxel. Anlotinib is a small molecule multi-targeted tyrosine kinase inhibitor that inhibits angiogenesis and tumor growth by targeting receptors such as VEGFR, PDGFR, FGFR, and c-KIT. Toripalimab is a monoclonal antibody immune checkpoint inhibitor targeting the programmed cell death protein 1 (PD-1) receptor to enhance anti-tumor immune responses. Nab-paclitaxel (nanoparticle albumin-bound paclitaxel) is a cytotoxic chemotherapeutic agent that stabilizes microtubules and inhibits cell division; its albumin-bound formulation improves drug delivery to tumors. The combination aims to synergistically inhibit tumor growth through antiangiogenic effects (anlotinib), immune activation (toripalimab), and direct cytotoxicity (nab-paclitaxel). This regimen has been investigated in phase II clinical trials for advanced or metastatic pancreatic cancer[1][6], with related studies in gastric cancer[2][4] and hepatocellular carcinoma[3].
