Overview

Annexin A2 short hairpin RNA (shRNA) is a sequence of RNA designed to silence the expression of the **Annexin A2 (ANXA2)** gene through the RNA interference (RNAi) pathway. Annexin A2 is a calcium-dependent phospholipid-binding protein that plays a critical role in cell surface fibrinolysis, cell-cell adhesion, and signal transduction. In the context of oncology, particularly **bladder cancer (BLCA)**, ANXA2 is often overexpressed and associated with high pathological grades, metastasis, and poor survival. By delivering shRNA—often via lentiviral or plasmid vectors—into target cells, the endogenous RNAi machinery (including the RISC complex) is recruited to degrade ANXA2 mRNA. This downregulation results in decreased cell proliferation, migration, and invasion, as well as a reduction in the secretion of proangiogenic factors and cytokines like IL-6, IL-8, and PDGF-BB.
