AnnuNovo is a proprietary, low-dose oral formulation of the mTOR inhibitor rapamycin (sirolimus) being developed by Annunovo, a subsidiary of Cambrian BioPharma. The drug is specifically designed to address ovarian aging and extend the female reproductive lifespan by delaying the depletion of the ovarian reserve. Mechanistically, AnnuNovo inhibits the mechanistic target of rapamycin complex 1 (mTORC1), which plays a critical role in the activation of primordial follicles. By slowing the rate at which these follicles are recruited into the growing pool, AnnuNovo aims to preserve ovarian function, delay the onset of menopause, and mitigate the health risks associated with the menopausal transition. The program is currently in the early stages of development, focusing on optimizing dosing to achieve tissue-specific effects in the ovaries while maintaining a favorable safety profile.
