Overview

Anrikefen (HSK21542) is a highly selective, peripherally acting kappa-opioid receptor (KOR) agonist developed by Haisco Pharmaceutical. It is designed to provide potent analgesia and anti-pruritic effects while minimizing the central nervous system (CNS) side effects typically associated with mu-opioid receptor agonists, such as respiratory depression, sedation, and potential for addiction. By selectively activating KORs on peripheral nerve terminals, anrikefen inhibits the transmission of pain and itch signals without crossing the blood-brain barrier. The drug is primarily being developed for the management of postoperative pain and the treatment of chronic kidney disease-associated pruritus (CKD-aP). Clinical studies, including those in elderly patients with obstructive sleep apnea (OSA), aim to characterize its efficacy and superior respiratory safety profile compared to traditional opioids.
