Anrikefon (HSK21542) is a peripherally restricted kappa-opioid receptor (KOR) agonist developed by Haisco Pharmaceutical Group. It is designed to provide potent analgesia and anti-pruritic effects by selectively activating KORs on peripheral sensory neurons, thereby avoiding the central nervous system (CNS) side effects—such as respiratory depression, sedation, and dysphoria—common to mu-opioid agonists and centrally acting KOR agonists. The drug is currently under clinical investigation for the management of postoperative pain and chronic kidney disease-associated pruritus (CKD-aP). Due to its peripherally restricted nature, it exhibits very low brain penetration, which enhances its safety profile compared to traditional opioid analgesics.
