ANT043 is an investigational anti-HER2 antibody fragment–drug conjugate (also termed an antibody fragment–drug conjugate, FDC) being developed by Antikor Biopharma for the treatment of HER2-positive solid tumors, including breast, ovarian, and gastric cancers. It uses a small single-chain antibody fragment (scFv-Fc) targeting the receptor tyrosine-protein kinase erbB-2 (HER2), coupled via Antikor’s OptiLink high–drug-to-antibody ratio technology (DAR approximately 6–10) to a cytotoxic payload, enabling rapid tumor penetration, efficient internalization, and potent picomolar cell-killing activity, including in trastuzumab-resistant models.[3][4][5][6][10][11] In multiple xenograft models, ANT043 has shown excellent tumor ablation and more rapid and complete tumor regression than a trastuzumab-based ADC, with superior tolerability and faster clearance from normal tissues, and its global development status is currently preclinical.[1][4][5][6][8][10][11]
