Overview

ANT2681 is a novel, specific, competitive metallo-β-lactamase (MBL) inhibitor developed for use in combination with the β-lactam antibiotic meropenem. Its primary mechanism of action is inhibition of MBLs—particularly New Delhi metallo-β-lactamase (NDM)—by interacting with the dinuclear zinc ion cluster in the active site of these enzymes, thereby restoring the antibacterial activity of meropenem against MBL-producing Gram-negative bacteria. ANT2681 has shown potent activity against NDM-positive Enterobacteriaceae and is being advanced into clinical development for serious infections caused by MBL-producing organisms, including complicated urinary tract infections (cUTI). The drug does not have intrinsic antibacterial activity but potentiates meropenem efficacy when used in combination[1][2][4][5][6][7].
