Antagomir-21 is a chemically modified antisense oligonucleotide designed to silence microRNA-21 (miR-21), a potent oncogenic and pro-inflammatory microRNA. Antagomirs are typically characterized by 2'-O-methyl modifications and a cholesterol conjugate at the 3' end to enhance cellular uptake and stability. By binding to miR-21, antagomir-21 prevents it from repressing its target mRNAs, such as PTEN, PDCD4, and SPRY2. This inhibition has been explored in various preclinical models for its potential to suppress tumor growth, reduce inflammation in autoimmune diseases like lupus and Crohn's disease, and promote tissue repair in conditions like spinal cord injury and acute kidney injury.
