Antagomir-301a is a synthetic, chemically modified oligonucleotide designed to inhibit the function of microRNA-301a (miR-301a). miR-301a is an oncogenic microRNA (oncomiR) that is significantly upregulated in several cancers, particularly pancreatic ductal adenocarcinoma, as well as breast, liver, and small cell lung cancers. It functions by directly targeting the 3' UTR of the CDKN1A (p21) tumor suppressor gene, leading to its downregulation and subsequent promotion of cell cycle progression and invasive potential. Treatment with antagomir-301a restores p21 levels, inducing G1 phase arrest and cellular senescence, thereby inhibiting tumor growth and metastasis. Research primarily conducted at institutions like the UT M. D. Anderson Cancer Center suggests its potential as a therapeutic strategy for pancreatic cancer.
