The anthrax and plague DNA vaccine is a multivalent biodefense candidate developed by the U.S. Army Medical Research Institute of Infectious Diseases (USAMRIID) to provide simultaneous protection against *Bacillus anthracis* and *Yersinia pestis*. The vaccine typically utilizes a combination of DNA plasmids, such as pV-PA (encoding the anthrax Protective Antigen) and pV-F1/V (encoding a fusion of the plague F1 and V antigens). The mechanism of action involves the delivery of these plasmids into host cells—often facilitated by a needle-free jet injector or gene gun—where the host's cellular machinery expresses the bacterial antigens. This process elicits a robust immune response, including the production of neutralizing antibodies that target the anthrax toxin and the plague bacterium's virulence factors. This DNA-based platform offers advantages in terms of stability, ease of manufacturing, and safety compared to traditional live-attenuated or protein-based vaccines.
