Anti-PD-1 antibodies are a class of immune checkpoint inhibitors that target the programmed cell death protein 1 (PD-1) receptor on T cells. By blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), these antibodies prevent the inhibitory signaling that tumors use to evade the immune system, thereby restoring T-cell mediated anti-tumor activity. This class includes several approved therapies such as pembrolizumab and nivolumab, and is widely used across various malignancies, particularly those with high microsatellite instability (MSI-H) or deficient mismatch repair (dMMR) status, including gastric and gastroesophageal junction adenocarcinomas.
