Anti-α5β8 integrin antibody is a monoclonal antibody designed to target and inhibit the α5β8 integrin complex. In the context of glioblastoma (GBM) research, this integrin has been identified as a critical component in the activation and release of Transforming Growth Factor-beta (TGFβ) within the tumor microenvironment. By blocking α5β8, the antibody prevents the immunosuppressive effects of TGFβ, thereby promoting the accumulation of germinal-center-like B cells and enhancing the anti-tumor activity of T cells. Preclinical studies in murine glioma models have shown that treatment with an anti-α5β8 antibody, particularly in combination with PD-1 blockade, can increase the infiltration of proliferating B-cells and significantly extend survival. This approach highlights a potential therapeutic strategy for modulating the immune landscape of 'cold' tumors like glioblastoma.
