Anti-αvβ8 mAb is a class of monoclonal antibodies that target the integrin αvβ8, a heterodimer of integrin subunits αv and β8. Integrin αvβ8 is a key activator of TGF-β signaling, binding to the latency-associated peptide (LAP) domain of latent TGF-β and triggering the release of active TGF-β1 and TGF-β3, which promotes an immunosuppressive tumor microenvironment. By blocking this interaction, anti-αvβ8 mAbs aim to reduce aberrant TGF-β signaling, reverse immunosuppression (e.g., by modulating macrophage polarization), and produce anti-tumor effects. Preclinical and clinical studies have shown tumor growth suppression and regression in models of solid tumors, such as squamous cell carcinoma, breast cancer, and colon cancer, both as monotherapy and in combination with checkpoint inhibitors (e.g., anti-PD-1/PD-L1). Clinical-stage candidates in this class include CRB-601 (developed by Corbus Pharmaceuticals) and PF-06940434 (developed by Pfizer).
