Overview

anti-βGal-hCD19 BiTE is a bispecific antibody construct designed in the Bispecific T-cell Engager (BiTE) format, consisting of two single-chain variable fragments (scFvs) linked in tandem. Unlike functional BiTEs which typically engage CD3 on T cells to induce cytotoxicity, this specific construct targets **human CD19** and **E. coli β-galactosidase (β-Gal)**. It is primarily utilized as a **negative control reagent** in preclinical studies to evaluate the activity of CD19-targeting T-cell engagers, such as blinatumomab. By binding to CD19 on target B cells without the ability to recruit T cells (due to the absence of a CD3-binding domain), it allows researchers to distinguish between biological effects caused by simple antigen binding and those resulting from T-cell-mediated cytotoxicity. This reagent is essential for demonstrating that the therapeutic mechanism of BiTE molecules is strictly dependent on the simultaneous engagement of both the tumor antigen and the T-cell receptor complex.
