**anti-5T4 CAR T cells** are genetically engineered T cells designed to express a chimeric antigen receptor (CAR) specific for the tumor-associated antigen **5T4** (trophoblast glycoprotein, TPBG), which is highly expressed on a variety of solid tumors, particularly ovarian cancer, and also on some acute myeloid leukemia (AML) cases[1][2][3]. The therapy involves isolating T cells from a patient or donor, transducing them ex vivo with a CAR construct utilizing a single-chain variable fragment (scFv) that targets 5T4, and re-infusing the cells to selectively recognize and kill 5T4-expressing cancer cells[1][3]. The mechanism of action is direct cell-mediated cytotoxicity against tumor cells by redirected T lymphocytes. Multiple CAR constructs have been described with different scFvs (e.g., H8, 2E4, H6), most utilizing co-stimulatory signaling domains such as 4-1BB and CD3ζ[1][3]. anti-5T4 CAR T cells have shown potent antitumor activity preclinically, selectively eradicating 5T4-positive tumors without affecting normal hematopoietic cells[2][3].
