The anti-ACBP monoclonal antibody is a research-stage biologic designed to neutralize extracellular Acyl-CoA Binding Protein (ACBP), also known as Diazepam Binding Inhibitor (DBI). ACBP is a phylogenetically conserved protein that, when secreted, acts as a hunger signal and a negative regulator of autophagy. In preclinical studies, this neutralizing antibody has demonstrated hepatoprotective effects in murine models of liver ischemia/reperfusion injury, acetaminophen toxicity, and nonalcoholic steatohepatitis (NASH) by inducing autophagy and reducing inflammation and fibrosis. In the context of B-cell lymphoma, ACBP is implicated in glucocorticoid resistance and immune evasion; the antibody aims to restore dendritic cell antigen presentation and enhance antitumor immunity by blocking the ACBP-GABA_A receptor-TSC22D3 axis.
