Drug intelligence / Profile preview
An anti-Activin A antibody is a therapeutic monoclonal antibody designed to bind and neutralize Activin A, a member of the transforming growth factor-beta (TGF-beta) superfamily. Activin A plays a critical role in various physiological and pathological processes, including bone remodeling, inflammation, and muscle wasting. In the context of the rare genetic disorder fibrodysplasia ossificans progressiva (FOP), mutations in the ACVR1 receptor cause it to abnormally perceive Activin A as an activating ligand, driving progressive heterotopic ossification (HO). By neutralizing Activin A, anti-Activin A antibodies block this aberrant signaling pathway, preventing ectopic bone formation and reducing painful tissue flare-ups. The leading clinical candidate in this class is Regeneron's garetosmab (Pasatru), which received FDA approval in August 2026 for the treatment of adults with FOP. Additionally, anti-Activin A antibodies are being explored in combination therapies for muscle preservation and fat loss in obesity management.
