Drug intelligence / Profile preview
An anti-activin A neutralizing antibody is a therapeutic monoclonal antibody designed to bind and neutralize activin A, a member of the transforming growth factor-beta (TGF-β) superfamily. Activin A plays a critical role in multiple pathological processes, including heterotopic ossification (as seen in fibrodysplasia ossificans progressiva, or FOP), tissue fibrosis (such as renal and pulmonary fibrosis), and muscle regulation. By blocking activin A from binding to its cognate type II receptors (ActRIIA/ActRIIB) and initiating downstream Smad2/3 signaling, these neutralizing antibodies inhibit aberrant osteogenesis, fibrotic remodeling, and muscle wasting. Garetosmab (REGN2477, brand name Pasatru) is a prominent clinical-stage example of this class, developed by Regeneron and approved by the FDA for FOP, as well as investigated in combination regimens for muscle preservation during weight loss.
