Overview

Anti-Activin E antibody is a first-in-class monoclonal antibody developed by iBio for the treatment of obesity and related cardiometabolic disorders. It targets Activin E, a hepatokine in the TGFβ family implicated in energy homeostasis and adipose tissue regulation. Human genetic studies show that loss-of-function mutations in INHBE (the gene encoding Activin E) are associated with reduced abdominal fat, lower triglycerides, and decreased risk of type 2 diabetes and cardiovascular disease. The drug works by binding to Activin E with high affinity, blocking its signaling pathway—resulting in fat-selective weight loss while preserving muscle mass. Preclinical studies demonstrate significant reductions in body fat as monotherapy and strong synergistic effects when combined with GLP-1 receptor agonists like semaglutide. The compound is engineered using AI-enabled epitope steering technology to overcome challenges associated with producing recombinant Activin E protein[1][2][3][4][6][7].
