Overview

Anti-ACVR2A antibodies are a class of monoclonal antibodies designed to selectively bind and inhibit the activin receptor type-2A (ACVR2A). ACVR2A is a transmembrane protein that serves as a primary receptor for ligands in the transforming growth factor-beta (TGF-β) superfamily, including activin A, activin B, and myostatin (GDF8). These ligands are potent negative regulators of skeletal muscle mass; thus, blocking ACVR2A signaling promotes muscle hypertrophy and increased lean mass. In the context of metabolic disease, ACVR2A inhibition has also been shown to reduce adipogenesis and improve fat metabolism. Recent development efforts, such as those by Biocytogen using their HUGO-eKO humanized mouse platform, have focused on creating fully human antibodies with high selectivity for ACVR2A over the closely related ACVR2B to avoid off-target effects like telangiectasia. These antibodies are being investigated for the treatment of obesity and sarcopenic obesity, particularly as a combination therapy with GLP-1 receptor agonists to preserve muscle mass during weight loss.
