An anti-ADAM17 antibody is a monoclonal antibody designed to target and inhibit A Disintegrin and Metalloproteinase 17 (ADAM17), also known as tumor necrosis factor-alpha converting enzyme (TACE). ADAM17 is a membrane-bound metalloproteinase responsible for the ectodomain shedding of various cell surface proteins, including TNF-alpha and epidermal growth factor receptor (EGFR) ligands (such as TGF-alpha and amphiregulin). Overexpression of ADAM17 is implicated in cancer progression, metastasis, and resistance to therapies like trastuzumab and EGFR tyrosine kinase inhibitors. By blocking ADAM17, these antibodies prevent ligand shedding, thereby suppressing downstream EGFR/HER receptor activation and tumor cell proliferation. Several preclinical candidates, such as D1(A12), D8P1C1, and MEDI3622, have been developed to evaluate this therapeutic strategy in solid tumors.
