anti-ADAM9-DM4 is a preclinical antibody-drug conjugate (ADC) developed through a collaboration between ImmunoGen and MacroGenics. It consists of a high-affinity monoclonal antibody targeting ADAM9 (A Disintegrin and Metalloproteinase 9) conjugated to the maytansinoid microtubule disruptor DM4 via a hindered disulfide sulfo-SPDB linker. ADAM9 is a cell surface protease overexpressed in various solid tumors, including lung, pancreatic, renal, and prostate cancers, where its expression correlates with poor prognosis. In preclinical studies, anti-ADAM9-DM4 demonstrated potent in vitro cytotoxicity across multiple ADAM9-positive tumor cell lines and induced significant tumor growth delay in xenograft models of non-small cell lung cancer (NSCLC).
