Overview

Anti-alpha-synuclein monoclonal antibodies are a class of target-specific immunotherapies designed to bind to alpha-synuclein (α-syn), a presynaptic neuronal protein that misfolds and aggregates into toxic fibrils (forming Lewy bodies) in synucleinopathies such as Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA). These antibodies aim to inhibit the cell-to-cell propagation of pathological alpha-synuclein, reduce neurotoxicity, and promote clearance of aggregates by microglia. Prominent clinical-stage candidates in this class include prasinezumab (developed by Prothena and Roche) and amlenetug (Lu AF82422, developed by Lundbeck).
