Anti-Annexin A2 monoclonal antibody is a therapeutic monoclonal antibody designed to target and neutralize Annexin A2 (ANXA2), a calcium-dependent phospholipid-binding protein that is highly expressed on the surface of aggressive cancer cells, particularly in triple-negative breast cancer (TNBC). Annexin A2 plays a critical role in tumor progression, invasion, and neoangiogenesis by binding to tissue plasminogen activator (tPA) and plasminogen, which accelerates the generation of plasmin in the tumor microenvironment. This localized plasmin generation subsequently activates matrix metalloproteinases, such as MMP-2 and MMP-9, leading to extracellular matrix degradation and tumor vascularization. By blocking Annexin A2, the monoclonal antibody disrupts this proteolytic cascade, significantly inhibiting tumor growth and angiogenesis. It has demonstrated long-term efficacy and a favorable safety profile in preclinical xenograft models of aggressive human breast cancer.
