Overview

Anti-AREG neutralizing antibody is a human monoclonal antibody designed to target and neutralize amphiregulin (AREG), a low-affinity ligand of the epidermal growth factor receptor (EGFR). AREG is typically released from the cell surface via TACE/ADAM17 protease cleavage and plays a significant role in driving EGFR phosphorylation, which promotes cell proliferation, migration, and differentiation. It is notably upregulated in advanced tumors and following chemotherapy, contributing to treatment resistance. By binding to AREG, the antibody prevents its interaction with EGFR, thereby inhibiting downstream signaling pathways. Preclinical research has demonstrated its potential to inhibit tumor growth and enhance chemotherapy efficacy in ovarian, breast, and prostate cancer models. Additionally, recent studies have explored its role in mitigating chronic inflammation and tissue fibrosis, particularly in models of colitis and Crohn's disease.
