**Anti-B7-H3 antibody** refers to a monoclonal antibody that specifically targets the immunoregulatory protein B7-H3 (CD276), a type I transmembrane glycoprotein overexpressed in many solid and some hematologic tumors with limited expression in healthy tissues[2][3][7]. B7-H3 contributes to tumor immune evasion by inhibiting T-cell activation, proliferation, and anti-tumor immune responses, and also supports tumor cell migration, invasion, angiogenesis, and chemoresistance[7]. The therapeutic mechanism of anti-B7-H3 antibody consists of binding selectively to B7-H3, blocking its inhibitory function on immune effector cells, and/or facilitating immune-mediated tumor cell killing (via ADCC, CDC, etc.)[7]. This antibody may serve as the basis for various drug formats: - Unconjugated monoclonal antibody as immunotherapy - Antibody-drug conjugates delivering cytotoxic payloads - Agents for radioimmunotherapy, bispecific T cell engagers, or CAR-T cell therapies targeting B7-H3[2][3][7] Many anti-B7-H3 antibodies are investigational, with several in clinical trials for cancers such as small-cell lung cancer, neuroblastoma, prostate cancer, and other advanced solid tumors[2][4][7].
