Overview

An anti-BAFF antibody is a monoclonal antibody designed to target and neutralize B-cell activating factor (BAFF), also known as B-lymphocyte stimulator (BLyS). BAFF is a member of the tumor necrosis factor (TNF) ligand superfamily and plays a critical role in the maturation, survival, and differentiation of B cells. By binding to BAFF, these antibodies prevent its interaction with its receptors (BAFF-R, TACI, and BCMA), thereby reducing the survival of autoreactive B cells and the production of pathogenic autoantibodies. This therapeutic approach is primarily utilized in the treatment of systemic autoimmune diseases. Belimumab was the first-in-class anti-BAFF antibody to receive regulatory approval for systemic lupus erythematosus (SLE). Other agents in this class, such as ianalumab and tabalumab, have been investigated for conditions including rheumatoid arthritis, Sjögren's syndrome, and various B-cell mediated disorders.
